Title: Development of a novel lipopolyplex platform for dual mRNA delivery by core- and surface-loading in the context of mucosal vaccination

Abstract: The recent success of mRNA-based vaccines has stimulated research into their potential use across diverse medical applications. However, as the field is still in its infancy, several limitations of lipid nanoparticles (LNPs), the current gold standard of mRNA delivery, need to be overcome. This thesis describes the development of a novel lipopolyplex platform to address key challenges of LNPs for mRNA delivery. By combining cationic and anionic polymers with cationic lipids, the core-shell nanoparticle supports independent dual nucleotide loading while achieving improved robustness during pharmaceutical processing. This design expands the potential use of mRNA therapeutics to new fields of applications, including inhalation-based medicine. As was found by the research described in this thesis, the localization of mRNA within the particle is critical: surface-loaded mRNA exhibited stronger expression compared to core-loaded mRNA, while expression kinetics were unaffected. Importantly, the LPPs demonstrated efficient co-delivery of multiple mRNAs to the same cell, which is particularly relevant for novel technologies such as CRISPR/Cas9. In this context, the transmembrane receptor CCR7 and chemokine CCL21 were explored as endogenous adjuvants for mucosal vaccination, demonstrating their promise as modulators of immune responses. Finally, the feasibility of LPPs with an oral delivery technology was investigated, showing successful formulation within polymer matrices and subsequent in vitro transfection.
Der Erfolg mRNA-basierter Impfstoffe zeigt das große Potenzial dieser Technologie in verschiedenen medizinischen Gebieten. Da sich die Technologie noch in ihren Kinderschuhen befindet, müssen Einschränkungen von Lipidnanopartikeln (LNPs), dem derzeitigen Goldstandard für mRNA-Transport, überwunden werden. Diese Arbeit beschreibt die Entwicklung einer neuen Lipopolyplex-Plattform, um zentrale Nachteile von LNPs zu adressieren. Durch die Kombination kationischer und anionischer Polymere mit kationischen Lipiden entsteht eine „Kern-Hülle“-Nanostruktur, die eine unabhängige, doppelte Nukleotid-Beladung ermöglicht und verbesserte Stabilität während pharmazeutischer Prozesse aufweist. Das erweitert die Einsatzmöglichkeiten von mRNA-Therapeutika zu neuen Anwendungsfeldern, einschließlich Inhalationstherapie. Unsere Untersuchungen zeigten, dass die Lokalisation der mRNA im Partikel entscheidend ist. Oberflächen-beladene mRNA erzielte stärkere Expression als Kern-beladene, bei unveränderter Expressionskinetik. Zudem gelang der Co-Transport von zwei mRNAs in dieselbe Zelle, besonders relevant für neue Technologien wie CRISPR/Cas9. Weiterhin wurden der Transmembran-Rezeptor CCR7 und das Chemokin CCL21 als potenzielle endogene Adjuvantien für Schleimhaut-Impfungen untersucht und zeigten vielversprechende immunomodulatorische Eigenschaften. Schließlich wurde die Eignung der Kombination von LPPs mit einer oralen Applikationstechnologie demonstriert, wobei die Formulierung in Polymermatrices die anschließende in vitro-Transfektion nicht negativ beeinflusste.
